POLISACÁRIDOS DE MACROALGAS ROJAS Y POLISACÁRIDOS / GLICOPROTEÍNAS DE MICROALGAS DE FUENTES NATURALES Y/O CULTIVOS: ESTUDIO ESTRUCTURAL, APLICACIONES INDUSTRIALES Y ACTIVIDAD BIOLÓGICA – MODIFICACIÓN QUÍMICA PARA MODULACIÓN DE PROPIEDADES
El objetivo del presente proyecto es estudiar los polisacáridos de macroalgas rojas, y los mucílagos extracelulares de especies de diatomeas en cultivo como modelos experimentales. La búsqueda de polisacáridos naturales o modificados químicamente se orientará a su aplicación como hidrocoloides en la industria alimentaria, excipientes de la industria farmacéutica o cosmética, o actividad farmacológica.
Dentro de las macroalgas, se completará el estudio estructural de los agaranos de plantas vegetativas de G. homersandii y se ha comenzado recientemente la caracterización de los sistemas de polisacáridos sulfatados de las algas Bryocladia thyrsigera y Asparagopsis taxiformis..
Los polisacáridos sulfatados se ensayarán como potencial materia prima para la formación de complejos interpolielectrolito, hidrogeles empleados en la liberación controlada de principios activos de interés farmacéutico o agronómico. Se someterán a reacciones de modificación química (cationización, sulfatación y desulfatación) a fin de modular sus propiedades biológicas y funcionales
Se está llevando a cabo la caracterización de los polisacáridos/glicoproteínas del mucílago extracelular de la diatomea Halamphora luciae aislada del estuario de Bahía Blanca. Dicho mucílago se separó en dos subfracciones: a) mucílago extracelular capsular o no soluble (fuertemente adherido a las paredes celulares) y b) mucílago coloidal o soluble (hidratos de carbono débilmente adheridos y el material soluble en el medio de cultivo). Se analizó la composición de los extractos y se determinará su homogeneidad. El estudio estructural se realizará por metilación, y espectroscopía de RMN de 1H y de 13C de los distintos productos purificados; de ser necesario se utilizarán técnicas bidimensionales.

The aims of this project are to study the polysaccharides from red macroalgae and extracellular mucilages from different diatom species obtained in cultures. The search for natural or chemically modified polysaccharides will be focused to their application as hydrocolloids in food industry, excipients for pharmaceutical and cosmetic industries and /or pharmacological activity.
Within the macroalgae, the structural analysis of the agarans from vegetative plants of G. homersandii will be completed and recently we started the characterization of the system of sulfated polysaccharides from Bryocladia thyrsigera y Asparagopsis taxiformis.
The sulfated polysaccharides will be assayed as potential raw material for the obtention of interpolyelectrolyte complexes, hydrogels employed for controlled release of active drugs of pharmaceutical and agronomic interest. Chemical modification reactions will be applied (cationization, sulfation and desulfation) in order to modulate their biological and functional properties.
The polysaccharides and/or glycoproteins isolated from the extracellular mucilage of the benthonic diatom Halamphora luciae are being characterized. The mucilage was separated in two subfractions: a) capsular extracellular non-soluble mucilage, which is strongly associated with the cell wall and b) colloidal or soluble mucilage, representing the fractions poorly attached to the cell wall and the material solubilized in the culture medium. The composition of the extracts was analyzed and their homogeneity will be determined. Structural determination of the different products will be carried out by methylation and 1H y de 13C NMR and if necessary by two dimensional NMR techniques.
